Abstract

En el presente trabajo se estudi6 la capacidad del lecho mesentérico de rata para producir y liberar prostanoides en presencia y en ausencia del endotelio vascular, así como los efectos de los agentes vasodilatadores acetilcolina (AC) y bradiquinina (BQ) sobre esa producción. Los prostanoides liberados se midieron por cromatografía líquida de alta presión (HPLC) . El lecho mesentérico perfundido produjo prostaglandina (PG) 6-cetoF 1a, tromboxano (TX) B2 (metabolitos estables de prostaciclina o PGI2 y TXA2 , respectivamente), PGE 2 y PGF2a . La remoción del endotelio produjo una reducción del 50% en la liberaci6n de PGI2 y de TXA2 . En el lecho intacto, la BQ 1 μ.M incrementó la liberación de PGI2 y de PGE2 , mientras que la AC 10 tM aument6 la liberación de PGI2 . En el lecho desendotelizado, la BQ incremento la producción de PGI 2 y la AC la de TX. La inhibición de la NO-sintetasa con L-NAME 100 pM inhibió el efecto de la BQ pero no modificó los efectos de la AC sobre la liberación de prostanoides. Se concluye que el lecho mesentérico de rata produce prostanoides vasodilatadores y vasoconstrictores de origen endotelial y extraendotelial, que la AC y la BQ estimulan la liberación de prostanoides de manera diferente y a través de mecanismos distintos y que el efecto de la BQ, especialmente en ausencia del endotelio, se traduciría en un mayor predominio de PG vasodilatadoras
