Engineering enhanced cellobiohydrolase activity

Abstract

Les cellobiohydrolases de la famille des glycosides hydrolases 7 (GH7 CBH) catalysent la dépolymérisation de la cellulose chez les eucaryotes cellulolytiques, ce qui en fait des cibles clés de découverte et d'ingénierie. Cependant, il reste un manque de relations robustes structure-activité pour ces cellulases d'importance industrielle. Ici, nous comparons les CBH de Trichoderma reesei (TrCel7A) et Penicillium funiculosum (PfCel7A), qui présentent une architecture multimodulaire composée d'un domaine catalytique (CD), d'un module de liaison aux glucides et d'un lieur. Nous montrons que PfCel7A présente des performances 60 % plus élevées sur la biomasse que TrCel7A. Pour comprendre la contribution de chaque domaine à cette amélioration, nous mesurons l'activité enzymatique pour une bibliothèque de chimères CBH avec des sous-domaines échangés, démontrant que l'amélioration est principalement causée par PfCel7A CD. Nous résolvons la structure cristalline de PfCel7A CD et utilisons cette information pour créer une deuxième bibliothèque de mutants TrCel7A CD, identifiant un double mutant TrCel7A avec une activité presque équivalente à PfCel7A de type sauvage. Dans l'ensemble, ces résultats révèlent des régions CBH qui permettent des améliorations ciblées de l'activité.

Las celobiohidrolasas de la familia de las glucósido hidrolasas 7 (GH7 CBH) catalizan la despolimerización de la celulosa en eucariotas celulolíticos, lo que las convierte en objetivos clave de descubrimiento e ingeniería. Sin embargo, sigue existiendo una falta de relaciones sólidas estructura-actividad para estas celulasas industrialmente importantes. Aquí, comparamos las CBH de Trichoderma reesei (TrCel7A) y Penicillium funiculosum (PfCel7A), que exhiben una arquitectura multimodular que consiste en dominio catalítico (CD), módulo de unión a carbohidratos y enlazador. Mostramos que PfCel7A exhibe un 60% más de rendimiento en biomasa que TrCel7A. Para comprender la contribución de cada dominio a esta mejora, medimos la actividad enzimática para una biblioteca de quimeras de CBH con subdominios intercambiados, lo que demuestra que la mejora es causada principalmente por PfCel7A CD. Resolvemos la estructura cristalina de PfCel7A CD y utilizamos esta información para crear una segunda biblioteca de mutantes de TrCel7A CD, identificando un mutante doble de TrCel7A con actividad casi equivalente a PfCel7A de tipo salvaje. En general, estos resultados revelan regiones de CBH que permiten mejoras específicas en la actividad.

Glycoside Hydrolase Family 7 cellobiohydrolases (GH7 CBHs) catalyze cellulose depolymerization in cellulolytic eukaryotes, making them key discovery and engineering targets. However, there remains a lack of robust structure-activity relationships for these industrially important cellulases. Here, we compare CBHs from Trichoderma reesei (TrCel7A) and Penicillium funiculosum (PfCel7A), which exhibit a multi-modular architecture consisting of catalytic domain (CD), carbohydrate-binding module, and linker. We show that PfCel7A exhibits 60% greater performance on biomass than TrCel7A. To understand the contribution of each domain to this improvement, we measure enzymatic activity for a library of CBH chimeras with swapped subdomains, demonstrating that the enhancement is mainly caused by PfCel7A CD. We solve the crystal structure of PfCel7A CD and use this information to create a second library of TrCel7A CD mutants, identifying a TrCel7A double mutant with near-equivalent activity to wild-type PfCel7A. Overall, these results reveal CBH regions that enable targeted activity improvements.
