Biotransformation of coumarins by Cunninghamella elegans

Abstract

La biotransformation microbienne de la coumarine (I) a été entreprise à l'aide d'une batterie de vingt-cinq micro-organismes. Parmi eux, le champignon Cunninghamella elegans NRRL 1392 était le seul microbe qui métabolisait efficacement la coumarine (I) en ombelliférone (II), 3, 4-dihydrocoumarine (V) et acide transcinnamique (VI). L'ombelliférone a également été biotransformée en 6, 7-dihydroxycoumarine (aesculetin, VII) et acide p-coumarique (VIII). D'autre part, la warfarine (III) a été biotransformée en 4`hydroxywarfarine (IX) et le dicoumarol (IV) a également été transformé en 4-hydroxycoumarine (X) par le même micro-organisme. Les structures des métabolites ont été établies en utilisant une seule propriété et des techniques spectroscopiques, y compris les points de fusion, la RMN 1 H, la RMN 13 C et la spectroscopie de masse. L'effet cytotoxique des substrats de coumarine et de leurs métabolites a également été étudié.

La biotransformación microbiana de cumarina (I) se llevó a cabo utilizando una batería de veinticinco microorganismos. Entre ellos, el hongo Cunninghamella elegans NRRL 1392 fue el único microbio que metabolizó eficientemente la cumarina (I) en umbeliferona (II), 3, 4-dihidrocumarina (V) y ácido transcinámico (VI). La umeliferona también se biotransformó en 6, 7-dihidroxicumarina (aesculetina, VII) y ácido p-cumárico (VIII). Por otro lado, la warfarina (III) se biotransformó en 4`hidroxi-warfarina (IX) y también el dicoumarol (IV) se transformó en 4-hidroxicumarina (X) por el mismo microorganismo. Las estructuras de los metabolitos se establecieron utilizando una sola propiedad y técnicas espectroscópicas que incluyen puntos de fusión, 1 RMN, 13 C RMN y espectroscopia de masas. También se investigó el efecto citotóxico de los sustratos de cumarina y sus metabolitos.

Microbial biotransformation of coumarin (I) was undertaken using a battery of twenty five microorganisms.Among them, the fungus Cunninghamella elegans NRRL 1392 was the sole microbe that efficiently metabolized coumarin (I) into umbelliferone (II), 3, 4-dihydrocoumarin (V) and transcinnamic acid (VI).Umbelliferone was also biotransformed into 6, 7-dihydroxycoumarin (aesculetin, VII) and p-coumaric acid (VIII).On the other hand, warfarin (III) was biotransformed into 4`hydroxywarfarin (IX) and also dicoumarol (IV) was transformed into 4-hydroxycoumarin (X) by the same microorganism.The structures of the metabolites were established using only one property and spectroscopic techniques including melting points, 1 H NMR, 13 C NMR and mass spectroscopy.The cytotoxic effect of coumarin substrates and their metabolites was also investigated.
