ผลของโซเดียมไนไตรท์และ/หรือ เมททิลพาราไธออนในขนาดที่ไม่ทำให้ปลาตายในปลาดุกพันธุ์ผสม (Clarias macrocephalus VS Clarias gariepinus)

Abstract

ศึกษาผลของโซเดียมไนไตรท์ และ/หรือ เมททิลพาราไธออนในขนาดที่ไม่ทำให้ปลาตายในปลาดุกพันธุ์ผสม แบ่งปลาทดลองออกเป็นกลุ่มควบคุม กลุ่มที่ได้รับโซเดียมไนไตรท์ (6.25-150 มก./ลิตร) เมททิลพาราไธออน (0.125-4.0 มก./ลิตร) และเมททิลพาราไธออนขนาด 0.25 มก./ลิตรร่วมกับโซเดียมไนไตรท์ (12.5-150 มก./ลิตร) หลังจากที่ปลาสัมผัสกับสารเคมีดังกล่าวนาน 24 ชั่วโมงแล้ว วัดสมรรถนะของเอนไซม์โฆลีนเอสเทอเรสทั้งในเลือดและสมอง ภาวะการเกิดเมทฮีโมโกลบินนีเมียและการเปลี่ยนแปลงค่าทางโลหิตวิทยา พบว่าสมรรถนะของเอนไซม์โฆลีนเอสเทอเรสทั้งในเลือดและสมอง ลดลงอย่างมีนัยสำคัญทางสถิติ (p<0.001) ในปลาดุกพันธุ์ผสมทุกกลุ่มที่สัมผัสกับเมททิลพาราไธออน เปอร์เซนต์การยับยั้งสมรรถนะของเอนไซม์โฆลีนเอสเทอเรสในเลือดและสมอง มีความสัมพันธ์กันและยังขึ้นอยู่กับความเข้มข้นของสารที่ได้รับ ค่าสัมประสิทธิ์ความสัมพันธ์ (r)=0.9671 เมททิลพาราไธออนไม่มีผลทำให้ค่าทางโลหิตวิทยาเปลี่ยนแปลง ในกลุ่มที่สัมผัสกับโซเดียมไนไตรท์ มีการสร้างเมทฮีโมโกลบินจากฮีโมโกลบินเพิ่มขึ้น อย่างมีนัยสำคัญทางสถิติ (p<0.01) โซเดียมไนไตรท์ไม่มีผลต่อสมรรถนะของเอนไซม์โฆลีนเอสเทอเรส ทั้งในเลือดและสมอง (p>0.01) ที่ความเข้มข้นตั้งแต่ 50 มก./ลิตร มีผลทำให้ค่าฮีมาโตคริตและฮีโมโกลบินลดลงอย่างมีนัยสำคัญทางสถิติ (p<0.05) จำนวนเม็ดเลือดแดงลดลงที่ความเข้มข้น 100 และ 150 มก./ลิตร (p<0.05) เมื่อศึกษาผลร่วมกันของเมททิลพาราไธออนขนาด 0.25 มก./ลิตร และโซเดียมไนไตรท์ที่ความเข้มข้นต่างๆ กัน (12.5-150 มก./ลิตร) พบว่าที่ความเข้มข้นของโซเดียมไนไตรท์สูงสุด ที่ให้ร่วมกับเมททิลพาราไธออน มีผลทำให้การเกิดเมทฮีโมโกลบินเพิ่มขึ้นอย่างมีนัยสำคัญ (p<0.05) เมื่อเปรียบเทียบกับกลุ่มที่ได้รับโซเดียมไนไตรท์อย่างเดียว ในขนาดเท่ากัน มีความแตกต่างกันของสมรรถนะของเอนไซม์โฆลีนเอสเมอเรส ทั้งในเลือดและสมองอย่างไม่มีนัยสำคัญทางสถิติ (p>0.05) ระหว่างกลุ่มที่ได้รับโซเดียมไนไตรท์อย่างเดียว กับกลุ่มที่ได้รับร่วมกับเมททิลพาราไธออนขนาด 0.25 มก./ลิตร และที่ความเข้มข้น 75-150 มก./ลิตร ทำให้ค่าฮีมาโตคริตและฮีโมลโกลบินลดลงอย่างมีนัยสำคัญ (p<0.05) นอกจากนั้น โซเดียมไนไตรท์ที่ขนาดตั้งแต่ 50 มก./ลิตร ยังทำให้จำนวนเม็ดเลือดแดงลดลงอย่างมีนัยสำคัญ (p<0.05) จากผลการศึกษาที่ได้นี้จะเห็นว่า สารเคมีทั้งสองตัวต่างก็มีความเป็นพิษต่อปลาดุกพันธุ์ผสม ความรุนแรงของการเกิดพิษจากโซเดียมไนโตรท์ต่อการเกิดภาวะเมทฮีโมโกลบินในเลือดอาจจะเพิ่มขึ้นได้เมื่อได้รับร่วมกับเมทิลพาราไธออน อย่างไรก็ตามมีความเป็นไปได้ที่เมททิลพาราไธออนอาจจะมีผลยับยั้งขบวนการรีดักชั่นที่เปลี่ยนเมทฮีโมโกลบินให้กลับไปอยู่ในรูปฮีโมโกลบินปกติได้
