Abstract

ขยะพลาสติกที่เพิ่มขึ้นในปัจจุบัน สามารถย่อยสลายหรือแตกตัวเป็นอนุภาคขนาดเล็กกว่า 5 มิลลิเมตร หรือไมโครพลาสติกจำนวนมากและส่งผลกระทบต่อสิ่งมีชีวิตในทะเล โดยมีการรายงานว่าไมโครพลาสติก polystyrene (PS) ที่อยู่ในสภาวะของทางเดินอาหารจำลองของสัตว์ไม่มีกระดูกสันหลังจะมีการปลดปล่อยสาร diethylhexyl phthalate ที่ออกฤทธิ์รบกวนระบบต่อมไร้ท่อและระบบสืบพันธุ์ ซึ่งในบริเวณชายฝั่งอ่าวไทยมีรายงานการปนเปื้อนไมโครพลาสติกชนิด polystyrene ในสัตว์ไม่มีกระดูกสันหลังหลายชนิด แต่ยังไม่มีการศึกษาผลกระทบของ polystyrene ต่อระบบสืบพันธุ์ของสัตว์ไม่มีกระดูกสันหลัง ในงานวิจัยนี้ได้เลือกใช้หอยแมลงภู่ Perna viridis ซึ่งเป็นหอยสองฝาที่มีคุณค่าทางเศรษฐกิจ และมีโอกาสได้รับไมโครพลาสติกผ่านการกรองกิน นำมาเลี้ยงในภาวะที่มีไมโครพลาสติกชนิด polystyrene แล้วตรวจสอบการสะสมและผลกระทบต่อระบบสืบพันธุ์ โดยนำหอยแมลงภู่จากแหล่งเพาะเลี้ยงใน อ.ศรีราชา จ.ชลบุรี มาเลี้ยงปรับสภาพที่สถานีวิจัยวิทยาศาสตร์ทางทะเลและศูนย์ฝึกนิสิต เกาะสีชัง จ.ชลบุรี ก่อนทดลองเลี้ยงในสภาวะที่มีเม็ด polystyrene ขนาด 0.6 - 1.0 มิลลิเมตร ที่ความเข้มข้น 0, 2 และ 20 mg/L เป็นเวลา 2 สัปดาห์ แล้วนำมาตรวจสอบการสะสมไมโครพลาสติกในทางเดินอาหาร โดยย่อยเนื้อเยื่อด้วยสารละลายโพแทสเซียมไฮดรอกไซด์ 10% แล้วนำสารแขวนลอยมาตรวจสอบปริมาณไมโคร พลาสติกภายใต้กล้องจุลทรรศน์แบบสเตอริโอ ผลการศึกษาไม่พบเม็ด polystyrene ขนาด 0.6 - 1.0 มิลลิเมตรในเนื้อเยื่อหอยแมลงภู่ แต่พบไมโครพลาสติกชนิดอื่นที่มีขนาดเล็กกว่า ซึ่งคาดว่ามีการสะสมอยู่ในหอยก่อนการทดลอง แสดงให้เห็นแนวโน้มว่าเม็ด PS ที่มีขนาดใหญ่กว่า 0.6 มิลลิเมตร ไม่สามารถเข้าไปสะสมในตัวหอยแมลงภู่ได้ และเมื่อนำอวัยวะสร้างเซลล์สืบพันธุ์มาเตรียมสไลดเนื้อเยื่อด้วยวิธี paraffin method ย้อมสี hematoxylin และ eosin นำมาตรวจสอบภายใต้กล้องจุลทรรศน์แบบใช้แสง พบว่าตัวอย่างทั้งหมดอยู่ในระยะที่ยังไม่มีการเจริญของเซลล์สืบพันธุ์ ข้อมูลที่ได้สามารถใช้เป็นแนวทางในการออกแบบการทดลองเพื่อตรวจสอบผลของไมโครพลาสติกต่อระบบสืบพันธุ์ของหอยแมลงภู่ต่อไป
