Characterization of a Cisplatin-resistant HeLa subline and mechanisms of cisplatin-resistance.

Abstract

ヒト口腔領域由来腫瘍細胞, UEDA, NAKATA, KANETSUKI, HSG, HSY, 子宮頸部由来HeLa, 外陰部由来A431の7種の培養細胞を用いてシスプラチンに対する感受性を検索した。腺癌細胞は扁平上皮癌細胞に比べてシスプラチンに対して高い感受性を示し, 唾液腺腺癌に化学療法が有用であると示唆された。また, 一方扁平上皮癌細胞は自然耐性を示した。シスプラチンに感受性の高い細胞は高い細胞内シスプラチン濃度を示し, シスプラチンの取り込みは殺細胞効果を反映すると考えられた。シスプラチン耐性のメカニズムを解明するためにHeLa細胞を用いてシスプラチン耐性細胞を分離した (HeLaDDP) 。同細胞はシスプラチン非存在下17ケ月間継代培養した後も約4.3倍の安定した耐性を発現していた。耐性細胞と母細胞との間で細胞内グルタチオン濃度に差はなかった。しかし, 耐性細胞はX線照射による生存率が高く, シスプラチンの取り込みは低下し, 放出に差はなかった。以上の結果から, シスプラチン耐性のメカニズムにはDNA修復能や取り込みの障害が関与していると考えられた。また, HeLaDDPはシスプラチン耐性機序を解明するにきわめて有用であると考えられた。
