Abstract

Vaksin konjugat dibuat dengan menggunakan bagian kapsul bakteri. Kapsul tersebut mengandung antigen polisakarida Vi, di mana Vi merupakan faktor untuk menentukan tingkat patogen di dalam bakteri dan antigen merupakan molekul struktur turunan dari bakteri yang dapat memicu respon imun protektif ketika dikombinasikan dengan molekul struktur yang lebih besar seperti carrier protein. Antigen polisakarida Vi berhubungan dengan imunogenisitas yang merupakan kemampuan suatu imunogen untuk menginduksi respon imun di dalam tubuh. Maka dari itu kadar Vi diuji untuk melihat dan menilai seberapa efektif dan protektif vaksin typhoid Vi jenis konjugasi ini. Metode HPAEC-PAD adalah kromatografi pertukaran anion berkinerja tinggi untuk kuantifikasi kandungan sakarida bebas dan total dari vaksin konjugasi tifoid (konsentrat bulk atau fill akhir) sakarida yang dipisahkan dideteksi dengan Pulsed Amperometric Dection (PAD). Metode tersebut perlu dilakukan validasi untuk menghasilkan data yang diyakini konsisten sesuai dengan spesifikasi yang telah ditetapkan. Tujuan penelitian ini adalah membuktikan bahwa metode HPAEC-PAD yang digunakan akan memberikan ketelitian, ketepatan serta ketegaran (Robustness) yang memenuhi persyaratan.
