MAS Yöntemiyle Domates Lekeli Solgunluk Virüsü’ne Dayanıklı Domates Hatlarının Geliştirilmesi

Abstract

Domates, Solanaceae familyasına ait dünyada ve ülkemizde yetiştiriciliği en fazla yapılan sebze türüdür. Domates yetiştiriciliğini olumsuz yönde etkileyen birçok hastalık etmeni ve zararlı bulunmaktadır. Domates lekeli solgunluk virüsü (Tomato spotted wilt virüs, TSWV), bazı ülkelerde karantina kapsamında bulunan, Thrips tabaci ve batı çiçek tripsi (Frankliniella occidentalis) tarafından taşınan tehlikeli bitki virüslerinden biridir. Bu amaçla, Ege Tarımsal Araştırma Enstitüsü domates gen havuzunda yer alan sofralık ve sanayilik tüketime uygun domates hattı hassas M68 ile Tomato Genetic Resource Center (TGRC)’den temin edilen Domates lekeli solgunluk virüsüne dayanıklı LA3667 ile melezleme yapılarak oluşturulan popülasyonda MAS yöntemi kullanılarak homozigot dayanıklı nitelikli hatların moleküler olarak seleksiyonu yapılmıştır. Bu amaçla F₂ ıslah kademesinde monogenik (1:2:1) açılımına göre 150 F₂ bireyinde 35 homozigot dayanıklı, 74 heterozigot dayanıklı ve 41 hassas birey elde edilmiştir. Dayanıklı olarak belirlenen homozigot dayanıklı hatlar, F₄ kademesine kadar kendilenmiş, F₄ kademesinde tekrar moleküler olarak testlenmiş, Sw-5 lokusu bakımından homozigot dayanıklı 3 hat ve 2 hassas domates ıslah hattı mekanik inokülasyon yoluyla biyolojik testlemeye alınmış ve dayanıklılık sonuçlarının, moleküler ve biyolojik olarak birbiriyle uyumlu olduğu görülmüştür.
