GENÉTICA PSIQUIÁTRICA E A ESTRUTURA PSICOPATOLÓGICA DO TRANSTORNO DEPRESSIVO MAIOR

Abstract

O transtorno depressivo maior (TDM) é um transtorno complexo comum com etiologia parcialmente genética. Estudos de associação genômica ampla (Gwas) e outros métodos genéticos de descoberta fornecem um meio para identificar mecanismos biológicos subjacentes a esse distúrbio comportamental que podem apontar para novos caminhos terapêuticos, aumentar as ferramentas de diagnóstico e produzir uma compreensão mais profunda da etiopatogenia desta condição. Estudos genômicos descobriram importantes genes associados a este distúrbio psiquiátrico que influenciam seus traços comportamentais e especificadores do transtorno. Desafios significativos permanecem na caracterização de influência específica de genes associados à doença e na priorização do acompanhamento funcional para torná-los úteis à compreensão da estrutura psicopatológica e desenvolvimento de terapêuticas. Em decorrência disso, são correlacionados dados genômicos que podem fornecer informações importantes sobre os mecanismos neurobiológicos, genéticos, epigenéticos e ambientais com um esforço no sentido de estabelecer quais aspectos da modulação biológica impactam sobre a psicopatologia do TDM. Descobertas e correlações descritas nesta revisão fornecem uma estrutura para aproveitar de forma eficiente os dados do projeto G-Was em apoio a fenótipos psiquiátricos clinicamente relevantes e direcionar para possíveis linhas de abordagem de tratamento.
