Uji Aktivitas Antioksidan Ekstrak Etanol 96% Daun Karamunting (Melastoma malabathricum L.) dengan Metode CUPRAC

Abstract

Radikal bebas merupakan senyawa yang mampu merusak struktur seluler di dalam tubuh, termasuk DNA, lipid, dan protein. Antioksidan merupakan senyawa yang dapat menghambat terjadinya proses oksidasi. Daun karamunting (Melastoma malabathricum L.) tumbuhan yang dapat dijadikan sebagai sumber antioksidan alami. Tujuan dari penelitian ini adalah untuk mengetahui kandungan metabolit sekunder yang terdapat pada ekstrak etanol 96% daun karamunting dan untuk mengetahui nilai EC50 yang diperoleh dari uji aktivitas antioksidan ekstrak daun karamunting diuji secara kuantitatif dengan metode CUPRAC (Cupric Ion Reducing Antioxidant Capacity). Daun karamunting (Melastoma malabathricum L.) diekstraksi menggunakan metode maserasi dengan pelarut etanol 96%. Pengujian aktivitas antioksidan daun karamunting dengan metode CUPRAC (Cupric Ion Reducing Antioxidant Capacity) dengan alat Spektrofotometer UV-Vis dengan pembanding kuersetin. Pengujian pembanding kuersetin yaitu dengan konsentrasi 1; 2; 3; 4; 5; ppm dan sampel ekstrak dengan konsentrasi 25; 50; 75; 100; 125; ppm. Pada pengujian skrining fitokimia mendapatkan hasil positif mengandung antioksidan pada uji alkaloid, fenol, flavonoid, saponin, steroid, tannin. Hasil penelitian EC50 dengan kuersetin yaitu 3.8059 ppm dan nilai EC50 ekstrak etanol 96% daun karamunting (Melastoma malabathricum L.) yaitu 61.6185 ppm. Kesimpulannya ekstrak etanol 96% daun karamunting tergolong aktivitas antioksidan yang kuat.
