TRIAGEM PARA ENZIMAS HIDROLÍTICAS DE FUNGOS FILAMENTOSOS ASSOCIADOS A SUBSTRATOS AGROINDUSTRIAIS

Abstract

Introdução: o isolamento de fungos a partir de substratos agroindustrias é uma alternativa para obtenção de micro-organismos produtores de enzimas hidrolíticas já adaptadas a esses substratos e, potencialmente, de interesse biotecnológico. Objetivos: isolar fungos filamentosos a partir de substratos agroindustriais e avaliar a atividade celulolítica e proteolítica. Material e métodos: milho triturado e soja (0,5g/substrato), adquiridas em mercado local, foram inoculados em meio Batata Dextrose Agar (BDA) suplementado com 0,1 μg/mL de cefalotina. As placas foram incubadas (25 ± 2°C) por até sete dias e, após o crescimento, os isolados foram purificados, caracterizados morfologicamente, agrupados em morfotipos e preservados (método de Castellani). Para verificação de atividade celulolítica (AC), representantes de cada morfotipo foram repicados em meio CMC (0,5% de carboximetilcelulose; 1,0g/L de glicose; 0,1g/L de extrato de levedura; 0,5g/L de peptona; 16,0g/L de ágar; água destilada q.s.p. 1,0 L), enquanto que para verificação de atividade proteolítica (AP) ocorreu a repicagem em meio BDA suplementado (9:1) com uma solução estéril de leite em pó desnatado constituído (10%). Para revelação da AC, utilizou-se solução aquosa de vermelho congo a 0,2% (15 min) e lavagem com 5,0 mL de solução de NaCl 1,0 M. O aparecimento de uma zona mais clara ao redor das colônias indicou AC positiva. Para revelação da AP, foi verificada a presença de um halo translúcido, que significa que o agente é proteolítico. Para linhagens com AC positiva, calculou-se o Índice de Relação Enzimática – IRE (D/d), em que D = diâmetro total (halo + colônia) e d = diâmetro da colônia. Resultados: fungos filamentosos foram obtidos a partir de ambos substratos (milho = 3,6 x 10² UFC/g, agrupados em cinco morfotipos; soja = 4,0 x 103 UFC/g, correspondendo a sete morfotipos). Com relação à AC, 100% dos isolados foram positivos, com IRE variando entre 1,02 a 3,00 (média ± desvio padrão = 1,47 ± 0,14). Com relação à AP de linhagens de milho, 40% (2/5) foram positivas. Conclusão: há uma grande variedade de fungos filamentosos associados a substratos agroindustrias, capazes de produzir enzimas hidrolíticas potencialmente úteis à bioindústria, que podem e devem ser explorados do ponto de vista biotecnológico.
