Uji Aktivitas Inhibitor Enzim Tirosinase Kombinasi Ekstrak Bunga Mawar (Rosa Damascena Mill) Dengan Fraksi Umbi Bengkuang (Pachyrizus Erosus)

Abstract

Enzim tirosinase adalah enzim yang bertanggung jawab atas terjadinya biosintesis melanin dalam pembentukan warna pigmen kulit dan penyebab hiperpigmentasi. Bunga mawar (Rosa damascena mill) dan umbi bengkuang (Pachyrhizus erosus) mengandung senyawa yang memiliki aktivitas inhibitor enzim tirosinase. Tujuan dilakukannya penelitian ini untuk mengetahui berapa besar aktivitas inhibitor enzim tirosinase dari ekstrak bunga mawar, fraksi umbi bengkuang, dan kombinasinya dengan perbandingan 1:1, 1:2, 2:1, 1:3 dan 3:1. Metode penelitian dilakukan dengan ekstraksi maserasi bunga mawar dengan etanol 70% dan ekstraksi umbi bengkuang dengan metode sokletasi dengan petroleum eter dan metanol, lalu difraksinasi cair-cair dengan etil asetat. Dari hasil ekstraksi diperoleh rendemen ekstrak bunga mawar 15,17% dan fraksi umbi bengkuang 12,5%. Hasil skrining fitokimia menunjukkan bahwa ekstrak etanol bunga mawar mengandung alkaloid, flavonoid, kuinon dan fenol, sedangkan fraksi umbi bengkuang mengandung alkaloid, flavonoid, saponin, fenol dan steroid. Pengujian aktivitas inhibitor enzim tirosinase menggunakan substrat L-tirosin dan kontrol positif asam kojak dengan pengukuran serapan menggunakan microplate reader pada panjang gelombang 492 nm. Hasil penelitian aktivitas inhibitor enzim tirosinase menunjukkan bahwa ekstrak bunga mawar memiliki nilai IC50 262,882 ppm dan fraksi umbi bengkuang dengan nilai IC50 43,148 ppm. Hasil penelitian inhibitor enzim tirosinase kombinasi ekstrak bunga mawar dengan fraksi umbi bengkuang dengan perbandingan 1:1; 1:2; 2:1; 1:3 dan 3:1 memiliki nilai IC50 secara berurutan yakni sebesar 26,598 ppm; 23,348 ppm; 29,880 ppm; 20,305 ppm dan 34,742 ppm.
