Starch-Gel Electrophoresis of Peanut Protein

Abstract

落花生蛋白質をトリスークェン酸緩衝溶液 (pH8.60) で抽出し, トリス-クエン酸及びホウ酸-水酸化ナトリウム (pH8.60) の不連続緩衝液系を用いて澱粉ゲル電気泳動を行ない次の結果を得た。(1) 尿素及びメルカプトエタノールを含まない澱粉ゲルでは幅の広い2個のバンドと不明瞭な2個のバンドが得られたが, 尿素濃度の増加と共にバンドの数は増加しかつシャープになった。特に5M尿素及び0.02Mメルカプトエタノールを含むゲルを用いると10数個のバンドが検出された。(2) 5M尿素及び0.02Mメルカプトエタノールを含むゲルを用いて34品種の落花生について電気泳動を行なったところ, 同一系統に属する品種の泳動パターンはそれぞれ同じであったが, バージニアと, スパニッシュ及びバレンシアとの間にパターンの差が認められた。(3) この差異はarachinの部分に存在し, conarachinの泳動パターンはすべての品種について同じであった。
