Conversion of D-phenylalanine to L-phenylalanine in humans.

Abstract

健康な女子2名にD-Phe-d5を8mg/kg体重, 他の必須アミノ酸および低タンパク質食とともに経口負荷し, 1, 4, 8時間後に採血した。血漿中の遊離フェニルアラニンをHPLCによりDL分割し, L画分について, Phe-d5/Phe-d0モル比をGC-MS-MIDにより測定した。この結果から, 血漿中のD-Phe-d5および, これから変換したL-Phe-d5の量を, 血漿中に天然に存在しているL-Phe-d0とのモル比として算出した。負荷後の血漿中のD-Phe-d5/L-Phe-d0モル比の最低は負荷1時間後の0.38 (被験者S: 0.16, T: 0.60の平均値), 最高は負荷4時間後の0.78 (S: 0.72, T: 0.84) であった。-Phe-d5/L-Phe-d0モル比の最低は負荷1時間後の0.015 (S: 0.014, T: 0.015), 最高は負荷4時間後の0.043 (S: 0.045, T: 0.040) であった。以上のごとく, 血漿中に遊離L-Phe-d5の存在が確認され, D-フェニルアラニンがL-フェニルアラニンに変換することが明らかとなった。
