Abstract

血小板/内皮細胞接着分子-1 (PECAM-1、CD31、以後ここでは“PECAM”と表すことにする) は、免疫グロブリンスーパーファミリーに属する130kDaの糖タンパク質である。この糖タンパク質は、血管内皮細胞間の接着部位に集積し、また血小板や末梢に存在する白血球の細胞表面には分散して存在している。また、骨髄や循環系に関係する多くの細胞に発現し、数種の細胞接着反応を仲介することができる。内皮細胞結合のような同じ細胞同士の接着 (同種細胞間接着) 及び内皮細胞への白血球の結合のような異なる細胞間の接着 (異種細胞間接着) に関わっている。分子レベルでは、PECAMは、同一分子同士でも結合 (PECAMとPECAMの結合) し、また、実験条件によってはある種の硫酸化グリコサミノグリカンがリガンドと思われる異分子間接着を行う。一方、白血球では、PECAMにリガンドや抗体などが結合すると、同じ細胞上のインテグリンファミリー接着分子が活性化されることが示されている。このように、PECAMは循環系の生理学や病理学において、様々な役割を担っている。
