Human Glycophorin A: Aspects of Glycosylation.

Abstract

グリコホリンA(GPA)は35kDaの糖タンパクで、ヒト赤血球上のシアリル化糖タンパクの中で最も豊富なものである(~0.2-1x106分子/赤血球)(1)。これは全配列が決定された最初の膜タンパクで131個のアミノ酸よりなり、今でも一度膜を貫通する型の膜タンパクの中で最も良く研究されたものの一つである(2、3)。GPAが糖化学者を惹き付けるのは、その細胞外ドメインに多くのグリコシドを持ちシアル酸含量が異常に高いためである(4-9)。ヒト赤血球グリコホリンのGPA遺伝子解析やその多型(10)、GPA抗原の構造と機能(1)に関する総説が最近いくつか出ている。本総説ではGPAのグリコシル化やグリコシル化サイト、GPAやその多型に存在する糖鎖の型、そして糖鎖のGPAの機能に果たす役割について調べた研究をまとめてみたい。さらに、グリコシル化サイトの同定、特にGPAやムチンに典型的な多くのO-グリコシドの結合したドメインの解析に関した技術の進歩についても述べたい(11)。
