Structure and Biology of Type VIII Collagen.

Abstract

VIII型コラーゲンは、ウシ大動脈内皮細胞の培養系における生合成産物として最初に報告されたので、従来は内皮細胞コラーゲン (EC) と呼ばれていた。このタンパク質は現在、トリおよび哺乳類のケラチノサイト、線維芽細胞、星状細胞腫や他の腫瘍細胞、さらに様々な大きさの血管に由来する内皮細胞などによって分泌されることが知られている。生体内ではVIII型コラーゲンの分布は限られており、例えば髄膜、骨膜、軟骨膜、角膜のデスメ膜などの細胞外マトリックスに存在する。また大小血管の内皮下層や内皮細胞の発芽培養系で形成される毛細血管様構造にも存在する。タンパク質のアミノ酸配列と核酸塩基配列から、VIII型コラーゲンはα1 (VIII) およびα2 (VIII) と名づけられた2本の異なるα鎖から構成されていることが明らかにされたが、細胞外の場所ではこれらのα鎖がどのような会合体を作っているのかまだよくわかっていない。しかしデスメ膜からタンパク質を抽出したところ、VIII型コラーゲンはこの組織においてヘテロ3本鎖構造をとっていることが示された。一方VIII型コラーゲンの生物学的機能に関する我々の知識はまだまだ不足している。VIII型コラーゲンはデスメ膜の特殊な六角格子構造の形成に関与していると考えられているが、発生分化の過程では本コラーゲンの発現は一定の期間、特定の場所に限られており、このことから組織再構築や分化にも関与していると思われる。
