Hyaluronan Synthase: New Directions for Hyaluronan Research.

Abstract

今日、ヒアルロン酸は脊椎動物の様々な組織やある種の連鎖球菌の莢膜に広く存在することが知られており、また変形性関節症の関節腔内に注入される補充物として医学応用されるなど、その重要性は年々増加している。ヒアルロン酸は特に、胚時期の細胞増殖や運動性の活発化している組織で、細胞外マトリックスの主要な構成成分として存在し、ヒアルロン酸結合分子や細胞表面受容体との相互作用により、細胞の接着、移動、分化をダイナミックに調節する。さらにこの遍在する高分子は形態形成や再生、創傷治癒、癌の浸潤・転移といった多彩な生命現象に関与することが示されている。他方、ヒアルロン酸の生合成機構解明に向けた研究、特にヒアルロン酸合成酵素を単離し同定する試みが、連鎖球菌や動物細胞を材料に過去十年以上にわたって精力的に行われてきた。ヒアルロン酸合成酵素を同定し、その遺伝子をクローニングすることは、ヒアルロン酸研究に新たな糸口を与えることになるだろう。近年我々をはじめ数グループが、ヒアルロン酸合成酵素の遺伝子を原核・真核細胞からクローニングし、ヒアルロン酸の生合成機構及び機能解析が急展開している。本レビューではヒアルロン酸合成酵素に関する最近の話題を紹介し、ヒアルロン酸研究が直面している問題点を指摘する。また最後に、ヒアルロン酸合成酵素遺伝子のヒアルロン酸生理機能研究への適用や臨床応用にも言及する。
