CONSERVACIÓN DE VAINILLA (Vanilla planifolia Jacks.) BAJO CONDICIONES DE LENTO CRECIMIENTO in vitro

Abstract

En México, la vainilla (Vanilla planifolia Jacks.) es considerada una especie endémica sujeta a protección especial debido a las severas afectaciones que ha sufrido su hábitat natural. En atención a esta problemática, es importante emprender programas para el rescate y conservación de este valioso recurso genético. El objetivo de este estudio fue evaluar el efecto de cuatro concentraciones (0, 10, 20 y 30 g L-1) de dos agentes osmóticos: manitol y polietilenglicol (PEG), y cuatro concentraciones (0, 1, 2 y 3 mg L-1) de dos inhibidores del crecimiento vegetal: ácido abscísico (ABA) y paclobutrazol (PAC), sobre la supervivencia y crecimiento in vitro de plantas de V. planifolia. En todos los tratamientos se utilizaron brotes de 0.5 cm de altura regenerados in vitro. Estos brotes fueron cultivados en medio de cultivo (MS). A los 180 d de cultivo se evaluó porcentaje de supervivencia, longitud de la planta, número de hojas, número y longitud de raíces. Los cultivos in vitro mostraron valores menores en las variables evaluadas de crecimiento, cuando fueron incrementadas las concentraciones de los agentes osmóticos e inhibidores en el medio de cultivo. Los tratamientos con PAC mantuvieron 100 % de supervivencia de los brotes. Sin embargo, este compuesto provocó la presencia de anormalidades en la parte apical y radical de las plántulas in vitro. Respecto al ABA, al utilizar 3 mg L-1 los brotes mostraron valores menores en todas las variables evaluadas y 90 % de supervivencia. Estos resultados permitieron establecer un método de conservación in vitro a mediano plazo de V. planifolia que prolonga el periodo entre subcultivos cada 180 d, sin afectar la viabilidad y fenotipo de las plantas.
