Abstract

Con el objetivo de determinar el pH óptimo al que cultivos celulares de cochinilla (Dactylopius coccus Costa) registran la máxima producción de ácido carmínico in vitro, se evaluaron varios niveles de pH (4.5, 5.0, 5.5, 6.0 y 6.5) y de fracciones de centrifugado (0, 2, 5 y 10 min) de células maceradas de embriones de cochinilla establecidos en medio nutritivo de Schneider. El contenido de ácido carmínico presente en cada muestra fue determinado mediante cromatografía líquida de alta resolución. La extracción del pigmento de las muestras se realizó con una solución de HCl 2M. Los límites de detección del pigmento fueron de 1.0 mg L-1 a 120.0 mg L-1. No se observó un efecto significativo del pH en el contenido de ácido carmínico ni en el número de líneas celulares, aunque el pH intermedio (5.5) tendió a dar mejores resultados.
