CONSTRUCCIÓN DE UN MAPA GENÉTICO PARA TRIGO HARINERO MEDIANTE RFLPs Y SSRs

Abstract

Una población de 117 líneas endogámicas recombinantes (RILs) F6, derivada de la cruza de las variedades Frontana e INIA66, se analizó mediante el uso de marcadores moleculares para construir un mapa genético que servirá de base para determinar los QTL de caracteres que segregan en la población. Un segundo objetivo fue determinar el potencial conjunto de la utilización de dos tipos de marcadores: RFLPs y los microsatélites o secuencias simples repetidas (SSRs), en el análisis genético de una especie de genoma complejo, como el que posee el trigo harinero (Triticum aestivum L. em. Thell.). Los progenitores se analizaron con 822 sondas de RFLPs y 68 pares de iniciadores. Mientras que los RFLPs mostraron un nivel bajo de polimorfismo (29 %), los SSRs tuvieron una mayor capacidad de detección del mismo (40 %). El análisis de las 117 RILs con 158 sondas RFLPs y 27 SSRs permitió la formación de 26 grupos de ligamiento. La especificidad de los SSRs fue un factor importante para la correcta designación de los grupos de ligamiento. Los resultados muestran que la combinación de diferentes clases de marcadores es una mejor estrategia para superar la limitante del nivel bajo de polimorfismo detectado al utilizar exclusivamente marcadores RFLPs en especies como el trigo.
