Abstract

La autofagia es un proceso omnipresente en todos los eucariotas, donde se ha descubierto una maquinaria genética adaptada para diferentes linajes celulares. Han sido descritos varios tipos de autofagia, involucrados en numerosos procesos fisiológicos donde funciona como un mecanismo protector; sin embargo, se ha descrito también que puede actuar en determinadas circunstancias como un mecanismo de muerte celular. En el proceso de diferenciación in vitro de Leishmania mexicana de la forma promastigote a la forma amastigote, hemos reportado un nuevo proceso selectivo parecido a autofagia dirigido a degradar complejos de membrana–microtúbulos redundantes, en respuesta a la elevación de la temperatura y acidificación del medio, imitando así los cambios que sufre el parásito durante la invasión del macrófago. Este proceso de diferenciación es importante para la sobrevivencia del parásito dentro del macrófago y la virulencia. Puede ser un modelo útil para estudiar el efecto de drogas dirigidas a bloquear la diferenciación y sobrevivencia del parásito, como nueva estrategia para combatir la enfermedad.
