Abstract

ウエルシュ菌のガス壊疽の病原因子であるα毒素(Cp-SMase)は,スフィンゴミエリン(SM)とホスファチジルコリン(PC)を分解する活性を有し,致死,壊死,そして,溶血活性を示す毒素である。セレウス菌のスフィンゴミエリナーゼ(Bc-SMase)は,致死,壊死活性を示さないが,リン脂質組成として,ホスファチジルコリンを含まず,スフィンゴミエリン(SM)を50%以上有するヒツジ赤血球に対して溶血作用を示す。Cp-SMaseは,細胞膜中でグリセロリン脂質の代謝亢進を惹起することによってラットの血管平滑筋の収縮やウサギ赤血球の溶血を引き起こすことが明らかにされてきたが,Bc-SMaseの作用機構に関しては不明であった。そこで,毒素作用とスフィンゴミエリナーゼ(SMase)活性の関係を解明するため,両者の作用を解析,比較した。その結果,1)Cp-SMaseとBc-SMasのヒツジ赤血球に対する溶血機構,さらに,2)Cp-SMaseの好中球やマクロファージに対する作用機構,および,3)Bc-SMaseの三次元立体構造解析と構造と機能の分析から,前者の作用は,SMase活性によるリン脂質の分解から始まりTrkA受容体およびGiを介する細胞内伝達系の活性化,そして,後者の作用は,細胞膜中に存在するSMのセラミドへの直接分解による膜の流動性低下により,それぞれの特徴的作用を示すことが明らかになった。従って,Cp-SMaseとBc-SMaseの作用は,いずれも,SMase活性によって細胞膜中のSM分解からスタートするが,その後の作用機構は,全く異なっていることが判明した。
