Abstract

極めて相同性の高い一次構造を持つ2種類の蛋白質が, 数個のアミノ酸残基の相違により全く異なった機能を持つという事実が Staphylococcus aureus の二成分蛋白毒素ロイコシジンならびにγヘモリジンにおいて明らかにされた。両者は, それぞれヒト白血球および赤血球上で孔を形成する蛋白毒素であるが, ロイコシジンの LukS 成分が, プロテインキナーゼA認識蛋白質であり, 本毒素成分のリン酸化と毒素活性特異性とに強い相関があることが明らかになった。また Panton-Valentine ロイコシジン遺伝子 (lukF-PVおよび lukS-PV) はφPVLと命名された S. aureus の溶原ファージゲノム上に存在した。本総説において, 次の5項目; (i) 二成分サイトリジンファミリー遺伝子の存在, (ii) lukF-PVおよびlukS-PVの溶原ファージゲノム上での存在, (iii) ロイコシジンとγヘモリジン成分の標的細胞への作用機構, (iv) ロイコシジンとγヘモリジン成分の細胞認識特異性を決定する最小領域, および (v) プロテインキナーゼA認識蛋白質としてのロイコシジンLukS成分, について今日までに明らかにされた研究成果を解説した。
