Abstract

D型ウエルシュ菌のe毒素は, 致死, 壊死作用を有するタンパク毒素で, 本菌によるヒツジの腸性中毒症の病原因子と考えられている。本研究では, e毒素の作用機構解明の手がかりを得るため, e毒素の in vivo と in vitro における作用, さらに構造と機能について検討した。まず, e毒素をラットに投与すると末梢血管の収縮による急激な血圧上昇が認められること, さらに本毒素は, in vitro で血管, 回腸などの摘出平滑筋組織を収縮させることを明らかにした。この作用は毒素が神経系にNa+の流入を促進し, その結果, 神経伝達物質が遊離, そしてCa2+チャンネルが活性化されて引き起こされることが判明した。一方, e毒素は脳組織において1種類のシアロ糖タンパク質に特異的に結合すること, e毒素の致死活性は中枢のドーパミン (DA) 代謝に影響を与える薬物で阻害されること, 毒素は脳内のDA量を減少させることから, 本毒素は, DA神経系に特異的に作用してDA分泌を促すと推察される。可逆的な化学修飾剤を用いる構造と機能に関するアプローチから, e毒素分子中のトリプトファンとチロシンの各1残基は毒素の受容体への結合に, そしてヒスチジン1残基は作用発現に関与していることが明らかとなった。本毒素は, 神経系に作用して神経興奮を引き起こす神経毒素の一つであると考えられる。
