Biological activities of chemically-treated glycolipids from Salmonella minnesota Re-mutant.

Abstract

Salmonella minnesota Re変異株よりフェノール:クロロホルム:石油エーテルで抽出した糖脂質(ReGl)をアルカリ(0.1N水酸化ナトリウムおよびアルカリ性NH2OH)あるいは弱酸(pH 4.5の酢酸ナトリウムおよび0.1N塩酸)で処理を行い,それぞれの生成物の生物活性を測定した。O-エステル結合脂肪酸の完全脱離したReGl-NH2OHはリムルス活性,鶏卵致死毒性および発熱毒性すべてが消失または大きく減少した。一方,ReGlから還元端のリン酸基とKDOが除かれたReGl-HClは鶏卵致死毒性と発熱毒性を失つたが,リムルス活性はそのままであつた。さらにReGl-NH2OHはマクロファージ活性化能およびリンパ球幼若化能も大幅に減少していたが,ReGl-HCIはマクロファージ活性化能を保持していた。ReGl-NaOHとReGl-pH 4.5は未処理のReGlとほとんど変らない上記の生物活性を示した。ラットのメチルコラントレン誘発腫瘍に対して,未処理のReGlとReGl-HClは約50%の治癒率を示すのに比較して,ReGl-NH2OHは延命効果を示すのにとどまつた。
