Purificación de proteínas de suero lácteo mediante coacervación con polielectrolitos: un estudio mediante el método de Monte Carlo

Abstract

El suero lácteo es el subproducto obtenido durante la elaboración del queso tras la separación del coágulo lácteo formado por la gelificación de las caseínas. Las proteínas del suero lácteo (PSL) constituyen el 0.7 % P/V de los nutrientes del suero lácteo y representan el 20% de las proteínas totales de la leche, siendo sus principales componentes β-lactoglobulina, α-lactoalbúmina y glicomacropéptido. Resulta de interés comprender el comportamiento a nivel macro de las proteínas presente en el suero lácteo. Utilizando un modelo de grano grueso para representar una molécula de β-lactoglobulina (BLG) y una cadena de polielectrolito (PE) se estudió la formación del complejo a distintos valores de pH. Se observó que el comportamiento del polielectrólito aislado es independiente de su pKa. Los resultados mostraron que la interacción entre BLG-PE es significativa a pH por debajo del punto isoeléctrico de la proteína ∼4.8. En este rango la carga de la proteína se ve modificada adquiriendo una mayor carga positiva lo cual se debe a la presencia de PE aniónico favoreciendo la interacción electrostática atractiva entre ambos. Por el contrario, a pH > 4.8 ambas macromoléculas tienen carga negativa, por lo tanto, la interacción entre las macromoléculas es de carácter repulsiva.
