Abstract

老人性骨粗鬆症についてカルシウム, リン等無機質の変化を捉えた研究は多いが, 有機質, ことに骨コラーゲンの変化を追求した研究は少なく, 老人性骨粗鬆症における骨コラーゲンの役割りについては不明の点が多い. この点を解明するための基礎研究として, 皮膚コラーゲンの架橋とリジン, ハイドロオキシリジンの関係, 低カルシウム食投与によるラットの実験的骨粗鬆症と骨コラーゲンのリジン含有量の関係を調べ興味ある所見を得たのですでに報告した. これによるとコラーゲンの架橋形成とともにリジン含有量が減少し, ハイドロオキシリジン含有量は変化しないことがわかった. また実験的骨粗鬆症の発現とともに骨コラーゲンのリジン含有量が増加し, 骨コラーゲンにおける架橋が減少していることがわかった.老人性骨粗鬆症を有する患者の骨についても骨コラーゲンに変化があるか否かを調べるために老人性骨粗鬆症を有する人とない人の大腿骨近位部の骨コラーゲンについてリジン含有量を林らの方法で, ハイドロオキシリジン含有量を Blumenkrantz の方法で定量した. それによると老人性骨粗鬆症を有する骨の骨コラーゲン内リジン含有量は平均31.5±0.33moles (コラーゲン105g当り) で正常の骨の骨コラーゲン内リジン含有量平均28.5±0.40moles に比べ増加している. ハイドロオキシリジン含有量は前者が平均3.98±0.20moles, 後者が平均3.57±0.30moles と両者の間に有意の差がなかった.実験的骨粗鬆症においても老人性骨粗鬆症においても骨コラーゲン内リジン含有量が増加していることから骨粗鬆症と骨コラーゲンの架橋との間に密接な関係があることがわかり, さらに骨コラーゲンの架橋と石灰化の関係を示す結果も得られたので考察を加えて報告する.
