The effect of bilirubin oxidase on serum bilirubin.

Abstract

Myrothecium verrucaria MT-1により産生されるビリルビンオキシダーゼ(EC 1,3,3,5)について,本稿ではin vivoおよびin vitroにおける本物質の酵素活性を確認した.血清ビリルビンはこの酵素によりビリベルジンとなり,その大部分はさらに水溶性ジアゾ反応陰性の酸化物に変化した.この酸化物の排泄経路を認べるために14Cビリルビンをビリルビンオキシダーゼにより酸化して14Cでラベルされた酸化物を作り,これをラットに静注し,尿・胆汁中の放射能を測定した.酸化物は尿・胆汁中に速やかに排泄され,特に尿中への排泄が多かった.安全性の確認が必要ではあるが,ビリルビンオキシダーゼは新生児黄疸等の減黄治療に応用し得ると考えられた.
