Abstract

Abstract. Skin is the outermost organ of the body that plays a role in the process of protection from UV radiation, which can cause the formation of free radicals and hyperpigmentation. Green tea leaves (Camellia sinensis (L) O. Kuntze) contain high polyphenol compounds, especially epigallocatechin gallate, which has potential as an antioxidant and tyrosinase inhibitor. This study aims to determine the antioxidant activity and tyrosinase inhibition in ethanol extract of green tea leaves. Ethanol extract of green tea leaves was obtained by reflux method using 96% ethanol solvent. Antioxidant activity test and tyrosinase inhibitor using DPPH method and dopachrome method based on enzymatic colorimetric testing. Antioxidant activity test results showed that green tea leaf extract has an IC50 value of 2.349 ppm, and has tyrosinase inhibitory activity with an IC50 value of 1000.422 ppm. Abstrak. Kulit merupakan organ tubuh terluar yang berperan dalam proses pelindungan dari radiasi sinar UV, yang dapat menyebabkan pembentukan radikal bebas dan hiperpigmentasi. Daun teh hijau (Camellia sinensis (L) O. Kuntze) mengandung senyawa polifenol yang tinggi khususnya epigallocatechin gallate yang berpotensi sebagai antioksidan dan inhibitor tirosinase. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan dan penghambatan tirosinase pada ekstrak etanol daun teh hijau. Ekstrak etanol daun teh hijau diperoleh dengan metode refluks menggunakan pelarut etanol 96%. Uji aktivitas antioksidan dan inhibitor tirosinase menggunakan metode DPPH dan metode dopakrom berbasis pengujian kolorimetri enzimatik. Hasil uji aktivitas antioksidan menunjukan bahwa ekstrak daun teh hijau memiliki nilai IC50 2,352 ppm, dan memiliki aktivitas penghambatan tirosinase dengan nilai IC50 1000,422 ppm.
