Abstract

Abstract. Incised wound healing requires effective topical therapy to prevent infection and accelerate tissue regeneration. However, long-term topical antibiotic use risks resistance and side effects. Arabica coffee contains bioactive compounds such as chlorogenic acid, caffeine, and polyphenols with antioxidant, anti-inflammatory, and antibacterial activities, making it a potential topical therapy alternative. This study aimed to compare the effectiveness of arabica coffee ointment with Neosporin on incised wound healing in male Wistar rats. This pure experimental study used a post-test only group design. Thirty male Wistar rats were randomly divided into three groups: negative control (no ointment), positive control (Neosporin), and treatment group (arabica coffee ointment). Wound healing evaluation was conducted on days 1, 3, 7, 10, and 14 through wound size measurement and assessment of wound color and surface condition. Repeated ANOVA analysis indicated significant differences between groups (p = 0.039). Wound color and surface condition assessments also showed better improvement in the coffee ointment group, with significant inter-group differences (p = 0.045). This study demonstrates that Arabica coffee ointment shows comparable effectiveness to Neosporin in wound healing based on wound size, and superior effectiveness in improving wound surface condition in the healing of incisional wounds in Wistar rats. Abstrak. Penyembuhan luka sayat memerlukan terapi topikal yang efektif untuk mencegah infeksi dan mempercepat regenerasi jaringan. Namun, penggunaan antibiotik topikal jangka panjang berisiko menimbulkan resistensi dan efek samping. Kopi arabika mengandung senyawa bioaktif seperti asam klorogenat, kafein, dan polifenol yang memiliki aktivitas antioksidan, antiinflamasi, dan antibakteri sehingga berpotensi sebagai alternatif terapi topikal. Penelitian ini bertujuan membandingkan efektivitas salep kopi arabika dengan neosporin terhadap penyembuhan luka sayat pada tikus Wistar jantan. Penelitian ini merupakan penelitian eksperimental murni dengan desain post-test only group design. Sebanyak 30 ekor tikus Wistar jantan dibagi acak ke dalam tiga kelompok, yaitu kontrol negatif (tanpa salep), kontrol positif (neosporin), dan kelompok perlakuan (salep kopi arabika). Evaluasi penyembuhan luka dilakukan pada hari ke-1, 3, 7, 10, dan 14 melalui pengukuran ukuran luka serta penilaian warna dan kondisi permukaan luka. Analisis ANOVA berulang menunjukkan perbedaan bermakna antar kelompok (p = 0,039). kondisi permukaan luka juga menunjukkan perbaikan lebih baik pada kelompok salep kopi arabika, dengan perbedaan signifikan antar kelompok (p = 0,045). Penelitian ini menunjukkan bahwa salep kopi arabika memiliki efektivitas yang setara dengan neosporin dalam penyembuhan luka berdasarkan variabel ukuran luka, serta lebih efektif dalam memperbaiki kondisi permukaan luka pada proses penyembuhan luka sayat tikus Wistar.
