Uji Daya Hambat Ekstrak Kopi Java Preanger terhadap Bakteri Cutibacterium Acnes

Abstract

Abstract. Resistance of Cutibacterium acnes (C. acnes) to the antibiotic minocycline is a serious problem in the treatment of acne vulgaris (AV). Java Preanger arabica coffee (Coffea arabica L) is rich chlorogenic acid and caffeine, can be used as a natural alternative for the treatment of AV. This study aims to compare the inhibitory power of Java Preanger Arabica coffee ethanol extract with minocycline as a first-line treatment for AV against C.acnes bacteria. This research uses an in vitro experimental method with well diffusion to measure the inhibition zone of C.acnes bacteria. Java Preanger Arabica coffee ethanol extract is made using the maceration method with concentrations of 75% and 100%. As a positive control, minocycline was used, while as a negative control, DMSO (Dimethyl Sulfoxide) was used. The bacterial inhibition zone was measured using a sliding period. The results showed that the ethanol extract of Java Preanger Arabica coffee at a concentration of 75% produced an average inhibition zone of 16.30 mm, while a concentration of 100% produced an inhibition zone of 10.18 mm. Minocycline produces the highest zone of inhibition, namely 28.27 mm. This shows that Java Preanger Arabica coffee extract is able to inhibit the growth of C. acnes bacteria, although its inhibitory power is lower than minocycline. (p-value = 0.0001) The mechanism of chlorogenic acid and caffeine by damaging protein synthesis by producing H₂O₂ thereby increasing bacterial membrane permeability and causing cell lysis. Abstrak. Resistensi Cutibacterium acnes (C.acnes) terhadap antibiotik minosiklin menjadi masalah serius dalam terapi acne vulgaris (AV). Kopi arabika Java Preanger (Coffea arabica L), kaya akan asam klorogenat dan kafein, bisa dijadikan sebagai alternatif alami untuk pengobatan AV. Penelitian ini bertujuan membandingkan daya hambat ekstrak etanol kopi arabika Java Preanger dengan minosiklin sebagai lini pertama untuk pengobatan AV terhadap bakteri C.acnes. Penelitian ini menggunakan metode eksperimental in vitro dengan difusi sumuran untuk mengukur zona hambat bakteri C.acnes. Ekstrak etanol kopi arabika Java Preanger disiapkan melalui metode maserasi dengan konsentrasi 75% dan 100%. Sebagai kontrol positif digunakan minosiklin, sedangkan kontrol negatif menggunakan DMSO (Dimetil Sulfoksida). Zona hambat bakteri diukur dengan menggunakan jangka sorong. Hasil penelitian menunjukkan bahwa ekstrak etanol kopi arabika Java Preanger pada konsentrasi 75% menghasilkan zona hambat rata-rata 16,30 mm, sedangkan konsentrasi 100% menghasilkan zona hambat 10,18 mm. Minosiklin menghasilkan zona hambat tertinggi, yaitu 28,27 mm. Hal ini menunjukkan bahwa ekstrak kopi arabika Java Preanger mampu menghambat pertumbuhan bakteri C.acnes, meskipun daya hambatnya lebih rendah dibandingkan dengan minosiklin. (p-value = 0,0001) mekanisme antibakteri asam klorogenat dan kafein adalah merusak sintesis protein dengan menghasilkan H₂O₂ sehingga meningkatkan permeabilitas membran bakteri dan menyebabkan lisis sel.
