Abstract

Abstract. Acne vulgaris is a skin abnormality in the pilosebaceous glands, characterized by the clinical appearance of blackheads, papules, pustules, and nodules that commonly occur in adolescents aged 12-25 years to adults aged 25-30 years. One of the causes of acne vulgaris is Propionibacterium acne bacteria. One of the plants that can be used to inhibit the growth of bacteria as an antibacterial is robusta coffee. This research aims to measure the size of the inhibitory zone of water extract compared to the ethanol extract of robusta coffee powder against Propionibacterium acnes using the disc diffusion method. This research uses an in vitro laboratory experimental study. Bacterial isolate P. acnes ATCC© 11827™ obtained from the Unisba Pharmacy laboratory followed by an inhibition test adapted to the McFarland 0.5 standard taken from the Unpad Teaching Hospital laboratory, with the independent variable of robusta coffee extract while the dependent variable was inhibition against Propionibacterium acnes. This research uses water extract and ethanol extract of robusta coffee, each made in 3 concentrations namely 50%, 75%, and 100%. The positive control is the antibiotic clindamycin, and the negative control is aquadest. The research results show no significant difference between the inhibitory power of water extract and ethanol extract against Propionibacterium acnes at concentrations of 50% and 75%. There is a substantial difference between the water extract and ethanol extract of robusta coffee at 100% concentration. Abstrak. Akne vulgaris merupakan kelainan kulit pada kelenjar pilosebasea, ditandai dengan gambaran klinis komedo, papula, pustula maupun nodul yang umum terjadi pada masa remaja usia 12 – 25 tahun hingga dewasa usia 25-30 tahun. Salah satu penyebab Akne vulgaris yaitu bakteri Propionibacterium acnes. Salah satu tanaman yang dapat dimanfaatkan untuk menghambat pertumbuhan bakteri tersebut sebagai antibakteri yaitu kopi robusta. Penelitian ini bertujuan untuk mengukur besar zona daya hambat ekstrak air dibandingkan ekstrak etanol bubuk kopi robusta terhadap Propionibacterium acnes menggunakan metode disc diffusion. Penelitian ini menggunakan studi eksperimental laboratorium secara in vitro. Isolat bakteri P. acnes ATCC© 11827™ didapatkan dari laboratorium Farmasi Unisba dilanjutkan uji daya hambat disesuaikan dengan standar McFarland 0,5 yang diambil dari laboratorium Rumah Sakit Pendidikan Unpad, dengan variabel bebas ekstrak kopi robusta sedangkan variabel terikat daya hambat terhadap Propionibacterium acnes. Penelitian ini menggunakan ekstrak air dan ekstrak etanol kopi robusta yang masing-masing dibuat dalam 3 konsentrasi yaitu 50%, 75%, dan 100%. Kontrol positif yang digunakan yaitu antibiotik klindamisin, kontrol negatif aquadest. Hasil penelitian menunjukkan tidak terdapat perbedaan yang bermakna antara daya hambat ekstrak air dan ekstrak etanol terhadap bakteri pada konsentrasi 50% dan 75%. Terdapat perbedaan yang bermakna ekstrak air dan ekstrak etanol kopi robusta pada konsentrasi 100%.
