Abstract

Die 4-Cumarat:Coenzym A Ligase 1 (4CL1) stammt aus dem pflanzlichen Sekundärstoffwechsel und gehört zur Familie der adenylat-bildenden Enzyme. Für strukturelle Untersuchungen des Reaktionsmechanismus der 4CL1 sollten Proteinvarianten produziert werden, die eine partielle 15N- Markierung mit der enzymvermittelten Peptid-Ligation ermöglichen. Die in der vorliegenden Arbeit hergestellten 4CL1 Varianten wurden, nach erfolgreicher Expression und Reinigung, auf ihre Kompatibilität mit der zur Peptid-Ligation verwendeten Trypsinvariante untersucht und im Vergleich mit der WT 4CL1 Variante durch CD- und Fluoreszenzmessungen sowie durch Aktivitätsmessungen genauer charakterisiert. Die Aminotransferase LivB ist an der Biosynthese des Aminoglykosid-Antibiotikums Lividomycin B in Streptomyces lividus beteiligt und katalysiert die Reaktion von 6′′′-Oxoparomomycin zu Paromomycin, einer Vorstufe von Lividomycin B. Die Kristallstruktur von LivB konnte mittels single wavelength anomalous dispersion (SAD) mit einer Auflösung von 1.5 Å aufgeklärt werden. Durch die Strukturaufklärung von LivB in Komplex mit dem Reaktionsprodukt Paromomycin konnten nähere Aussagen zum Reaktionsmechanismus von LivB getroffen werden.

The 4-coumarate coenzyme A ligase 1 (4CL1) belongs to the secondary metabolism of plants and is a member of the adenylate forming enzyme superfamily. For structural studies of the reaction mechanism protein variants should be produced which allowed partial labelling with 15N using enzyme mediated peptide ligation. The 4CL1 variants produced in the presented work were, after successful expressing and purifying them, checked for their compatibility to the trypsin variant used for peptide ligation, they were furthermore characterised through CD and fluorescence measurements as well as activity tests in comparison to WT 4CL1. The aminotransferase LivB is involved in the biosynthesis of the aminoglycoside antibiotic lividomycin B in Streptomyces lividus and catalyses the reaction of 6′′′-oxoparomomycin to paromomycin, a precursor of lividomycin B. The crystal structure of LivB could be solved to a resolution of 1.5 Å using single wavelength anomalous dispersion (SAD). Structure determination of LivB in complex with the reaction product Paromomycin allowed a closer look at the possible reaction mechanism of LivB.
