In der vorliegenden Arbeit wurde eine naїve scaffold Bibliothek, basierend auf dem humanen γB-Kristallin (hγBC), gegen die N-terminale, extrazelluläre Domäne des Parathyroidhormon-1-Rezeptors (nPTH1R), einem Vertreter der Klasse B GPCRs, selektiert. Vier Selektionsrunden (biopanning) wurden durchgeführt, wobei eine Variante (2G4) wiederholt isoliert werden konnte. Der Rezeptor wird von ihr in einer 1:1 Stöchiometrie mit niedrig mikromolarer Affinität gebunden. Zwei Phenylalanine an den Randomizierungspositionen 2 und 17 scheinen den größten Anteil am Bindungsereigniss zu haben, zumal die restlichen 7 umliegenden Aminosäuren ebenfalls ihren Teil zur Interaktion der beiden Moleküle beitragen. Eine Kompetition der Bindung von PTH an nPTH1R durch 2G4 und umgekehrt konnte nicht nachgewiesen werden, was auf unterschiedliche Epitope beider nPTH1R-Interaktionspartner schließen lässt. Potentielle Anwendungen für diesen vielversprechenden Binder könnten zum einen in Ko-Kristallisationsexperimenten, oder aber auch in der Affinitätschromatographie zur verbesserten Homogenität des Rezeptors aus Zellextraktionen liegen.
In this thesis, a naive scaffold library, based on human γB crystallin (hγBC), was selected against the N-terminal extracellular domain of parathyroid hormone 1 receptor (nPTH1R), a class B GPCR. Four rounds of a biopanning process were performed and one hγBC binding variant, called 2G4, was repeatedly isolated. It binds the nPTH1R with low micromolar affinity in a 1:1 stochiometry. Two phenylalanines on randomization positions 2 and 17 were found to contribute the most to the binding interaction and the surrounding seven residues also participate in the binding event. There is no competition binding of 2G4 and PTH to nPTH1R via identical or overlapping binding sites. This promising binder could have several potential applications, such as in co-crystallization experiments or in affinity chromatography to increase the homogeneity of the receptor.
