Influence of cellular dsRNA binding proteins in the replication process of a dsRNA virus

Abstract

Die Fragestellung dieser Arbeit war, ob Proteine des Virus der infektiösen Bursitis (IBDV) mit den aviären Proteinen p68 und NF45 (nuclear factor 45) interagieren. Beide Hühnerproteine wurden exprimiert und gereinigt um spezifische Seren herzustellen. Beide Proteine ändern ihre nukleäre Lokalisation nach einer Infektion mit IBDV zu einer zytoplasmatischen. Dort wurde die Kolokalisation von NF45 mit dem viralen Ribonukleoprotein VP3 und der RNA-abhängigen RNA Polymerase VP1 und von p68 mit der viralen Protease VP4 mit konfokaler Laserscan Mikroskopie detektiert. Durch Co-immunpräzipitation wurde geziegt, dass NF45-VP3 und p68-VP4 direkt interagieren. Die Kolokalisation beider Paare war abhängig von der Zeit, aber unabhängig vom Virusstamm, von Interferonen und von der infizierten Spezies. Bei verminderter Expression des zellulären Proteins durch siRNA konnten höhere Virustiter gemessen werden. Dies ist ein Hinweis, dass sowohl NF45 als auch p68 antivirale Eigenschaften besitzen.
