Die Chlorogensäure:Glukarsäure-Kaffeoyltransferase aus Solanum lycopersicum

Abstract

Aus Keimlingen der Tomate (Solanum lycopersicum) wurde ein Enzym isoliert, welches als Chlorogensäure:Glukarsäure-Kaffeoyltransferase(CGT) klassifiziert wurde und die Synthese von Kaffeoylglukarsäure und Kaffeoylgalaktarsäure aus Chlorogensäure katalysiert. Die im Zuge dieser Arbeit isolierte cDNA-Sequenz codiert für ein Protein mit einer Länge von 380 Aminosäuren und einem putativen Signalpeptid von 24 Aminosäuren für den ER-Import und anschließende Sekretion. Die Funktionalität von SlCGT wurde durch heterologe Expression in Hefe und Tabak bewiesen. Analysen der SlCGT-Proteinsequenz ergaben die Klassifizierung des Proteins als GDSL-Lipase-ähnliches Protein. Proteine dieser Familie besitzen eine katalytische Triade aus Ser-Asp-His. Allerdings wiesen Mutagenesestudien darauf hin, dass die abgeleitete putative katalytische Triade nicht als katalytisches Zentrum in die Acyltransferase-Reaktion der CGT dient. Die SlCGT ist damit das erste Beispiel eines GDSL-Lipase-ähnlichen Proteins, das seine hydrolytische Aktivität verloren und eine völlig neue Funktion im pflanzlichen Sekundärstoffwechsel entwickelt hat.
