Abstract

Im Rahmen dieser Arbeit wurden 79 Naturstoffe aus Pflanzen, Pilzen und Flechten hinsichtlich evtl. Einflüsse auf die Thrombozytenfunktion untersucht. Es stellte sich heraus, dass nur einer dieser Naturstoffe, das Purpurogallin, eine signifikante Hemmung der Aggregation nach Stimulation mit Thrombin auslösen kann. Weiteres Interesse galt dem Permethylpurpurogallin, der methylierten Form des Purpurogallins. Es zeigte sich, dass keine der nachgewiesenen Wirkungen des Purpruogallins auf das Permethylpurpurogallin übertragen werden kann. Der erstmalige Nachweis der Polo-Like-Kinase 1 ist im Rahmen dieser Arbeit gelungen. Ebenfalls konnte nachgewiesen werden, dass die PLK-1 in Thrombozyten in phosphorylierter Form vorliegt. Da ein Zusammenhang zwischen der PLK-1 und den MAPKinasen im Rahmen des Primings der PLK-1 beschrieben wird, haben wir auch untersucht, ob sich die Aktivität der MAPKinasen durch Purpurogallin hemmen lässt. Es konnte nachgewiesen werden, dass Purpurogallin einen hemmenden Einfluß auf die Aktivität auf die MAPKinasen ERK und p38 in Thrombozyten hat.

Within the scope of this work, 79 natural products from plants, mushrooms and lichens were examined with regard to possible effects on the platelet function. It turned out, only the purpurogallin, can trigger a significant inhibition of aggregation after stimulation with thrombin. Another drug was the permethylpurpurogallin, the methylated form of purpurogallin. It was shown that none of the detected effects of purpruogallin can be transferred to the permethylpurpurogallin. The first-time detection of polo-like kinase 1 has been successful in this work. It has also been shown that PLK-1 is present in platelets in phosphorylated form. As a link between the PLK-1 and the MAPKinases is described in the context of the priming of PLK-1, we also investigated whether the activity of the MAPKinases can be inhibited by purpurogallin. Purpurogallin has been shown to have an inhibitory effect on the activity on MAPKinases ERK and p38 in platelets.
