Abstract

Two types of intrinsically disordered proteins were investigated in this thesis to scrutinise their aggregation behaviour. First, the temperature-driven inverse phase transition of thermoresponsive peptide polymers was studied as a function of the amino acid sequence with electron paramagnetic resonance spectroscopy. The findings of this chapter will help to precisely tune the biophysical properties of thermoresponsive peptide polymers for the desired applications in biotechnology and medicine. Second, the systematic investigation of the interaction of myelin basic protein (MBP) with the single lipids of the cytoplasmic side of the myelin sheath revealed the importance of the synergy of the lipids to ensure MBP's physiologic role for the compaction of the myelin sheath and may further the understanding of pathological processes related to neurodegenerative diseases.4207 intrinsisch unstrukturierte Proteine; thermoresponsive Peptid-Polymere; inverser Phasenübergang; Elektronenspinresonanz-Spektroskopie; basisches Myelinprotein; Protein-Lipid-Wechselwirkung; Lipid-Monoschichten; Sphingomyelin

In dieser Arbeit wurde das Aggregationsverhalten zweier verschiedener Arten intrinsisch unstrukturierter Proteine untersucht. Zum einen wurde der Temperatur-abhängige inverse Phasenübergang von thermoresponsiven Peptid-Polymeren als Funktion der Aminosäuresequenz mit Elektronenspinresonanz- Spektroskopie untersucht. Die Erkenntnisse dieses Kapitels unterstützen die präzise Einstellung gewünschter biophysikalischer Eigenschaften für vorgesehene Anwendungen in Biotechnologie und Medizin. Zum anderen erfolgte die systematische Untersuchung der Wechselwirkung des basischen Myelinproteins (MBP) mit den einzelnen Lipiden der zytoplastischen Lipidschicht des Myelins. Diese zeigte die Bedeutung des Zusammenwirkens der Lipide für die physiologische Rolle des MBPs, die Kompaktheit der Myelinscheide sicherzustellen. Dies erlaubt Rückschlüsse für die Aufklärung pathologischer Prozesse im Zusammenhang mit neurodegenerativen Erkrankungen.
