Matric Potential and Overburden Potential in Gels for Plant Tissue Culture.

Abstract

本論では, 寒天とゲランガムをゲル化剤としたゲルに起因するマトリックポテンシャルについて検討した.その結果, 通常の植物組織培養で用いられるゲル化剤濃度におけるマトリックポテンシャルは, どちらのゲルでも-0.lkPa程度であり, 全水ポテンシャルに対する割合は無視できること, および培養体への水分供給の支配要因には成りえないことを明らかにした.さらに, ゲルに加えられた荷重の圧力は, 通常のゲル化剤濃度においては, 全量が荷重ポテンシャルとなり, ゲル内部の水圧を増加させることが分かった.また, ゲルからの“離しょう”が, ゲル内の水分平衡過程における排水現象として説明づけられた.しかし, ゲル培地の透水係数が極めて小さいという事実を考えると, 完全な水分平衡に至るまでには長時間を要すると考えられ, 離しょうを含むゲル培地中の水移動のフラックスを検討するには, 水移動の抵抗と関係する透水係数も含めて考えなければならない (Fujiwara, 1991) ,今後はゲル中の透水係数の変化, さらには溶質や酸素の拡散について検討を行う予定である.
