Obtención y caracterización de quitina biológica y su desacetilación a quitosano

Abstract

Tradicionalmente la obtención de la quitina y su derivado principal el quitosano implica la DM (desmineralización), DP (desproteinización) y desacetilación química del desperdicio de camarón, utilizando ácidos y bases fuertes en altas concentraciones, lo que provoca la depolimerización y la pérdida de otros productos de valor agregado tales como proteínas y pigmentos. Por otra parte, en este trabajo se evaluaron cuatro proteasas comerciales (Protamex, HT Proteolitic, Deterzyme y Proteasa N) en la obtención de quitina. Para este propósito, las enzimas comerciales se incubaron individualmente con quitina parcialmente purificada obtenida de una FAL (fermentación ácido láctica) del desperdicio de camarón. La Proteasa N mostró mayor actividad sobre la quitina. Una vez seleccionada la proteasa se evaluaron diferentes niveles de pH, temperatura y concentración de enzima. Los mejores resultados fueron obtenidos a un pH 7.5 a 40°C con una concentración de enzima de 0.5% (p/p). Bajo estas condiciones se obtuvo hasta un 99.3% de DP, recuperando quitinas biológicas con un 0.85 ± 0.16% de contenido de proteína. El proceso de desproteinización enzimática de la quitina es un método económicamente atractivo, ya que evita altos gastos de energía, agua y reactivos. En conjunto con la FAL se tiene un tratamiento biológico para la purificación total de la quitina, evitando que esta sufra daños en su estructura, al mismo tiempo que se busca obtener quitinas con mínimos residuos de proteína. La quitina obtenida por desproteinización enzimática fue caracterizada y desacetilada químicamente para obtener quitosano que a su vez fue caracterizado.
