Abstract

La senescencia celular es un fenómeno en el que las células cesan su proliferación en respuesta a un número limitado de duplicaciones o a un estrés que induzca senescencia en forma prematura. Este fenómeno está asociado al envejecimiento y a múltiples enfermedades asociadas a la edad como cáncer y enfermedades neurodegenerativas. Estudios actuales han demostrado que las células senescentes secretan citocinas, quimiocinas y factores de crecimiento con un perfil proinflamatorio que pueden contribuir al deterioro causado por dichas patologías. Al conjunto de éstas moléculas secretadas se le conoce como Fenotipo Secretor Asociado a la Senescencia (SASP, por sus siglas en inglés) y se han identificado diversos componentes, tales como la IL-6, IL-7, IL-8, TNF-, MCP-2, MIP-3, GRO y HGF, entre otros. En este trabajo, se indujo Senescencia Prematura Inducida por Estrés (SIPS, por sus siglas en inglés), Senescencia Prematura Inducida por Inhibición del Proteosoma (SPIIP) y Senescencia Replicativa (SR) en un cultivo de fibroblastos de pulmón de ratón y se caracterizaron mediante los parámetros clásicos de senescencia: Detención de la proliferación y sobreexpresión de p16 y -Galactosidasa. Además, se evaluaron los componentes del SASP en un estudio comparativo entre los 3 tipos de senescencia. Los resultados obtenidos muestran que la expresión de los marcadores clásicos de senescencia es idéntico en los 3 tipos estudiados, más no así el SASP, el cual disminuyo notablemente para la SPIIP, sugiriendo que el SASP es dependiente del estímulo que induzca la senescencia.
