OPTIMASI STERILISASI EKSPLAN PADA KULTUR IN VITRO BAMBU PETUNG (Dendrocalamus asper)

Abstract

Bambu petung (Dendrocalamus asper) merupakan salah satu jenis bambu yang banyak digunakan karena strukturnya kokoh dan kuat. Tingginya permintaan bambu petung mengakibatkan perlunya metode alternatif untuk perbanyakan bibit bambu petung agar tidak mengalami kelangkaan. Salah satu metode yang dapat digunakan adalah kultur in vitro. Metode ini dipilih karena mampu menghasilkan bibit tanaman dalam jumlah banyak dengan sifat genetik seragam dalam waktu relatif singkat. Namun dalam penerapannya, kontaminasi masih menjadi kendala utama kultur in vitro dan dapat menghambat produktifitas. Optimasi sterilisasi eksplan menjadi salah satu faktor penting dalam mengurangi kontaminasi serta tidak menghambat pertumbuhan eskplan. Penelitian ini bertujuan untuk mengetahui jenis sterilan, konsentrasi sterilan dan durasi perendaman optimal untuk mengurangi kontaminasi dan mendukung pertumbuhan eksplan pada kultur in vitro bambu petung. Penelitian dilakukan dengan perlakuan tunggal menggunakan jenis dan konsentrasi sterilan alkohol (50% dan 70%), (NaOCl pada clorox 50% dan 100%), dan (fungisida 1, 2 dan 3 g/L) dengan variasi durasi perendaman (0, 10, 20, 15, dan 30 menit). Berdasarkan tingkat kontaminasi dan waktu munculnya kontaminan, alkohol 70%, NaOCl dalam klorox 100% dan fungisida 2 g/L merupakan sterilant tunggal terbaik. Kombinasi sterilan NaOCl pada clorox 100%-15 menit dan fungisida 2g/L-20 menit merupakan sterilan kombinasi terbaik dalam mengurangi tingkat kontaminasi jamur yaitu sebesar 33,33% dan mendukung pertumbuhan tunas pada 6 HST dengan persentase 33,33%. Berdasarkan hasil penelitian dapat disimpulkan bahwa kombinasi sterilan NaOCl pada clorox 100%-15 menit dan fungisida 2g/L-20 menit merupakan sterilan yang optimal dalam mengurangi tingkat kontaminasi serta mendukung pertumbuhan eskplan dalam kultur in vitro bambu petung.
