A Novel HSF1-mediated Death Pathway

Abstract

熱ショック転写因子 (heat shock transcription factor, HSF) は,一群の熱ショック蛋白質 (heat shock protein, Hsp) の発現制御に関わる転写因子群として知られ,哺乳動物細胞では3種類のHSF (HSF1, HSF2, HSF4) が明らかにされている.このうち,HSF1は蛋白質の変性を感知し,Hspの発現制御において中心的な役割を担い,温熱ストレスをはじめとする様々なストレスに対する耐性の獲得を通して細胞生存に働いている.一方,我々は活性型HSF1トランスジェニックマウス,ならびに HSF1欠損マウスの解析から,HSF1が細胞死を導く働きがあることを示唆する結果を得ていた.本稿では,HSF1がHspとは全く逆の働きを持つ細胞死促進因子TDAG51 (T-cell death associated gene 51) を直接誘導することを示し,これまで知られていた細胞防御能だけでなく細胞死誘導能を併せ持っていることを明らかにした.さらに,このHSF1-TDAG51経路が生体内でも重要な機能を担っていることを明らかにした.この経路と蛋白質変性にともなう細胞運命の決定との関わりについて概説する.
