Abstract

培養歯根膜細胞を移値りることにより, 歯根膜とセメント質を再生させるという新しい治療法を検討する第一段階として, 近親交配ラットを実験群, 対照群, 歯根移植群に分けて本実験を行った。実験群は, ラット歯根膜細胞をin vitroで培養し, それをラットの象牙質片上に播種し, さらに培養を行い細胞を増殖させた後, ラット頭蓋に形成した骨欠損部に移植した。一方, 対照群は培養歯根膜細胞を付着させていない象牙質片を移植し, 歯根移植群は新鮮な歯根膜が付着した象牙質片を移植し, 組織学的観察を行った。その結果, 実験群では2週後の6例中1例に, 4週後の10例中4例に, 頭蓋骨とは連続していないセメント質様硬組織の形成がみられたが, 6週後には全面にアンキローシスが生じていた。対照群では2週後に, 増殖した骨とアンキローシスが一部に生じており, 4, 6週後には全てにアンキローシスの所見がみられた。歯根移植群では, 2週後には歯根膜が残存し4週後には一部象牙質に吸収がみられ6週後には全てにアンキローシスが生じていた。これらの結果より, 培養歯根膜細胞移植により歯根膜, セメント質を再生できる可能性が示唆された。しかし, 実験群は, 新鮮な歯根膜を移植した歯根移植群と同様に, 6週後にはアンキローシスがみられたことから, 培養歯根膜細胞を移植する場合に, 適切な時期に咬合力あるいはそれに類似した機能を与えることが必要ではないかと考えられた。
