An improved method for comprehensive investigation of subgingival microflora with a particular reference to obligate anaerobic spirochetes.

Abstract

本研究は歯周病の臨床検査として偏性嫌気性スピロヘータを含めた歯肉縁下プラークの細菌叢を検索する方法を確立するという目的で行った。被験者28名において歯周ポケット内細菌叢を検索し各培地・培養法の比較, 検討を行った。非選択培地としてはスチールウールジャー法を用いたEG寒天培地が適していることが分かった。また偏性嫌気性スピロヘータの培養については, プレートインボトル法を用いた10%Rumen液入りMedium10培地が優れているということが示された。以上の2つの培地に10種類の選択培地を加え患者と健常者について歯周ボケット内の細菌叢の検索を行ったが, 特にスピロヘータは半数以上の患者において総菌数の10%以上培養によって検出され, これは位相差顕微鏡の鏡検におけるスピロヘータの割合と同様の値になった。本研究に用いた検索法により歯周ポケット内の偏性嫌気性スピロヘータを他の細菌と同叢に培養・検索することが可能になった。
