Toxicidade aguda da dipirona sódica in vitro utilizando o bioindicador de toxicidade Artemia salina Leach.

Abstract

Estima-se que a automedicação atinja cerca de oitenta milhões de brasileiros e a dipirona é um dos analgésicos mais utilizados, entretanto esse fármaco pode causar conhecidas reações adversas como agranulocitose, nefrite intersticial, hepatite, alveolite, pneumonite e doenças cutâneas graves como síndromes de Seteves-Johnson e de Lyell, sendo motivo de restrição desse fármaco em muitos países. Dessa forma, o presente estudo visa analisar através de um estudo do tipo experimental, com abordagem quantitativa e descritiva, avaliar a toxicidade e identificar a concentração Letal 50% (CL50) da dipirona sódica gotas referência e da genérica, ambas disponíveis comercialmente e produzidas no Brasil, comparando-as entre si. Realizou-se o ensaio de letalidade em Artemia salina de acordo com a metodologia descrita por Meyer (1982), com algumas modificações. Com a realização do teste de toxicidade em Artemia salina, para a dipirona genérica, a CL50 obtida após 24 horas foi de 986,9 µg/mL. Já a dipirona de referência apresentou CL50 de 654,1 µg/mL. Diante da análise da presente pesquisa, constatou-se que a dipirona de referência apresentou indícios de toxicidade em naúplios do microcrustáceo Artemia salina apresentando uma CL50 ≤ 1000µg/mL. A dipirona genérica apresentou uma CL50 ≤ 1000µg/mL com intervalo acima de 1000 ug/mL indicando atoxicidade para essa amostra.
