Mechanism of viral antigen processing and presentation.

Abstract

Zinkernagel & Doherty (1974)によるキラーT細胞のウイルス感染標的細胞認識におけるMHC拘束性の発見から20年足らずで, この問題がウイルス抗原プロセッシングに関するMHC拘束性を反映したものであることが明らかになってきた。この経過も興味あるもので, この間にクラスI及びクラスII経路の違いも明らかになってきた。従来, 熱不活化インフルエンザウイルスを用いて抗原提示機構の独自の研究を続けてきた我々の研究室では, 国産のMHC表現欠損細胞をいただいて, ウイルス抗原提示機構の研究の新しい局面を切り開いている。インフルエンザウイルス-L929細胞 (H-2k) の系で研究を続けてきた経過から, 変異細胞はH-2kマウス由来のものである必要があった。C3Hマウス由来の Methylcholanthrene 誘導繊維肉腫細胞でウイルス感染によっては (内因性抗原提示) 抗原提示は出来ないが, NP抗原エピトープを細胞外から加えた時は (外因性抗原提示) 抗原提示出来ることがわかった。本MHC表現欠損細胞は, トランスポーター産生とクラスI分子産生と両者に欠損のある新しいタイプの特異細胞であることがわかった。そしてこのような原因による免疫不全症があればINF-γ投与により回復することが示唆された。
