Abstract

従来タンパク質のリン酸化は, ラジオアイソトープ標識法やウエスタンブロット法を用いて研究されてきたが, 我々は質量分析法による高感度分析法の有効性について基礎的な検討を行った. まずリン酸化部位が既知の標準ペプチドやカゼインを用いて, Post-source decay モードによるMALDI-TOF質量分析を行い, セリンおよびトレオニンリン酸化ペプチド検出における有効性を確認した. しかしチロシンリン酸化ペプチドには, 必ずしも有効ではないこともわかった. 検出感度を上げるために, 市販のリン酸化ペプチドおよびリン酸化タンパク質濃縮キットの使用方法の最適化を行い, 有効性を確認した. さらに我々はこの方法をラットミエリンのミエリン塩基性タンパク質の分析に応用し, リン酸化ペプチドの高感度検出と, 新しいリン酸化部位の特定に成功した.
