Abstract

Un phage filamenteux portant la séquence peptidique TLTTKLY a été isolé à partir d'une banque de présentation de phages heptapeptidiques contre un virus de la maladie de Newcastle vélogénique (NDV). Afin d'étudier le potentiel de ce phage spécifique en tant que réactif immunologique dans le pathotypage du virus, une méthode basée sur un dosage immuno-enzymatique (ELISA) a été développée. Cette méthode permet de différencier les souches vélogéniques des souches mésogéniques et lentogéniques. Un test de liaison à l'équilibre en solution a montré que les interactions entre le phage et toutes les souches de NDV donnaient lieu à deux constantes de dissociation (Kdrel) très différentes. Sur la base des premières valeurs de Kdrel, les souches NDV peuvent être classées en deux groupes ; le premier comprend les souches vélogéniques et le second comprend les souches mésogéniques et lentogéniques. Ces résultats indiquent un degré élevé de corrélation entre les affinités de liaison et le pathotypage des souches de NDV à l'aide du phage TLTTKLY.

Se aisló un fago filamentoso que portaba la secuencia peptídica TLTTKLY de una biblioteca de presentación de fagos heptapéptidos contra un virus velogénico de la enfermedad de Newcastle (NDV). Con el fin de investigar el potencial de este fago específico como reactivo inmunológico en la patotipificación de virus, se desarrolló un método basado en el ensayo inmunoabsorbente ligado a enzimas (ELISA). Este método puede diferenciar las cepas velogénicas de las cepas mesogénicas y lentogénicas. Un ensayo de unión en equilibrio en solución mostró que las interacciones entre el fago y todas las cepas de NDV dieron lugar a dos constantes de disociación muy diferentes (Kdrel). Con base en los primeros valores de Kdrel, las cepas de NDV se pueden clasificar en dos grupos; el primero comprende las cepas velogénicas y el segundo consiste en las cepas mesogénicas y lentogénicas. Estos resultados indican un alto grado de correlación entre las afinidades de unión y la patotipificación de las cepas de NDV utilizando el fago TLTTKLY.

A filamentous phage bearing the peptide sequence TLTTKLY was isolated from a heptapeptide phage display library against a velogenic Newcastle disease virus (NDV). In order to investigate the potential of this specific phage as an immunological reagent in virus pathotyping, an enzyme-linked immunosorbent assay (ELISA)-based method was developed. This method can differentiate the velogenic strains from the mesogenic and lentogenic strains. An equilibrium-binding assay in solution showed that the interactions between the phage and all the NDV strains gave rise to two widely differing dissociation constants (Kdrel). Based upon the first Kdrel values, NDV strains can be classified into two groups; the first comprises the velogenic strains, and the second consists of the mesogenic and lentogenic strains. These results indicate a high degree of correlation between the binding affinities and pathotyping of NDV strains using the TLTTKLY phage.
